Induced pluripotent stem cells are extensively used for in vitro modelling of human diseases, drug discovery and regenerative medicine. However, to fully understand the potential of stem cell based strategies we must first know the cellular processes essential for stem cell function. Mitochondria, the organelles involved in the cellular energy production, have been proposed to play a key role in the stem cell biology. Interestingly, mitochondria contain their own genome, the mitochondrial DNA (mtDNA), which is essential for the mitochondrial respiration. The aim of this project is to gain novel mechanistic insights into mtDNA maintenance and metabolism in human stem cells. It is largely unknown how mtDNA is replicated in stem cells and how the mtDNA levels and integrity are regulated during differentiation. This study will help to better understand the role of mitochondria in stem cells and advance the potential of stem cell based strategies.
