Schizophrenia is a severe psychiatric disease. Its mechanisms are not known. Brain diseases can be modelled using induced pluripotent stem cells. Previous iPSC studies have failed to find altered gene expression inschizophrenia because of the genetic heterogeneity. We minimize the disease-irrelevant noise in iPSC-derived cells by studying clinically well-characterized monozygotic twin pairs discordant for schizophrenia. We identify genetic factors associated with the risk of schizophrenia twins, and genetic factors contributing to the actual illness. We also investigate the impact of inflammation. Preliminary data show that brain cells contribute to risk and onset of schizophrenia, and that different pathways are involved in women and men. The potential prevention and cure will ultimately reduce human suffering and socioeconomic burden of SCZ. The project offers seeds for renewed clinical practices and boosts commercial drug development and diagnosis.
