Depressive and anxiety disorders are very common and often have a developmental origin. Early risk factors such as parental malcare, neglect or abuse predispose individuals to later disease. They lead to high stress exposure in early life and a resulting maldevelopment of networks in the brain regulating emotional behaviour. Most consistent abnormalities have been found in the function of the amygdala and prefrontal cortex after early adversity. These brain regions are particular vulnerable to early-life insults due to their ongoing development during childhood and adolescence. Our research aims to unravel the cellular and circuit mechanism that underlie abnormal amygdala and prefrontal cortex functional development by early-life stress. This may confer the increased risk to depressive and anxiety disorders later in life. Our results will help to develop new strategies for disease prevention as well as for better treatment.
