Considering the significant plant diseases potyviruses cause, we need to understand potyviral pathogenesis and virus-host plant interactions to address the challenges in production of healthy plants. We study potato virus A (PVA, a potyvirus) infection including how it multiplies, defends itself against hosts’ antiviral actions and forms virus particles. We have observed that a large protein complex follows PVA RNA throughout the infection cycle and suggested that it is required to safeguard the integrity of PVA RNA. We shall work towards the high-resolution structure of this complex. We aim in understanding how this complex and the cellular localization of PVA RNA regulate viral gene expression. We shall study the mechanism of particle formation and compare the tip-structure found from one end of potyviral particles to the RNA-bound complex to compare their relationship with each other. Knowledge on how to prevent stabile particle formation will enable resistance breeding.
