Drug delivery to the brain and central nervous system is limited by efflux transporters in the blood-brain barrier. The two main efflux transporters recognize a wide variety of drug substrates and are therefore key obstacles in delivering drugs to the brain. Genetic variation can affect the functionality of the transporters, and our hypothesis is that drugs that are normally prevented from entering the brain by these transporters would be able to reach much higher brain concentrations in patients with transporter variants that have low activity. Our aim is thus to establish how genetic variation in these efflux transporters impact drug exposure, and evaluate if this can be used to improve drug treatment in adults. Our studies will also generate data that helps us understand the molecular basis for substrate recognition and transport, which is a second aim of this project.
