Bile acid (BA) transporters are essential in maintaining the balance of potentially toxic BAs, their salts and conjugates in the human liver. Disturbances in the amounts and activities of BA transporters due to e.g. hepatic diseases, genetic variation and drug inhibition may cause severe liver damage. The biological basis for transporter regulation and such adverse reactions is not well understood. The aim of the project is to identify and study the presently poorly known factors that regulate the amounts, trafficking and functions of BA transporters on cellular membranes. The project utilizes the state-of-the-art hepatocyte models and techniques, and the results will be translated to analyze BA homeostasis in patients with hepatic diseases. The work is supported by a high-quality group of national and international collaborators. The results will provide a better understanding of how BA homeostasis is controlled and new tools to predict BA-dependent hepatic toxicities.
