Biophysical signaling plays a major role in the maintenance of all cellular functions and is associated with the progression of several diseases. Physical changes in the cell microenvironment alter the homeostasis of tissues through yet poorly characterized mechanisms. In this project we aim in understanding how physical alterations impact the functional differentiation and maintenance of bilayered tubular mammary epithelia. How does the multidirectional mechanosignaling, especially Ca2+-dependent signaling, between luminal and myoepithelial cells and their stroma impact this process and how is this altered along cancer progression. In these studies we utilize more physical methods combined with state-of-the-art imaging and an active international collaborative network. While these studies will shed a light on the mechanobiology behind normal and transformed mammary epithelia, they can also reveal general mechanisms behind the homeostasis of tubular epithelial tissues.
