Our cells contain compartments, or organelles, in which many crucial cellular reactions occur. When our organelles are not assembled correctly, severe disease can be the result. We are currently focusing our attention upon the peroxisome, an organelle involved in lipid breakdown and several other important cellular functions. It is not fully understood how cellular components are targeted to peroxisomes, but increased knowledge of this process will contribute to studies of peroxisomal disease and to other technological applications. We propose a state-of-the art approach that will simultaneously examine the ability of thousands of different protein variants to travel to, and be inserted within, the peroxisomal membrane. By this analysis, we will conclusively determine what specific properties of a targeting sequence allow proper peroxisomal delivery. In addition, we will seek, then carefully investigate the function of, novel factors involved in peroxisome assembly.
