Type 1 diabetes (T1D) is a common autoimmune disease typically manifesting in childhood. T1D occurs as a consequence of an immune-mediated destruction of the insulin-producing ß-cells in the pancreas. The pathogenesis of T1D is currently poorly understood, and therefore no effective ways to prevent the onset of T1D currently exist. Autoantibodies produced by B lymphocytes are currently the best biomarker available to predict future risk of T1D. In this project, we will analyze specialized subsets of T lymphocytes (T follicular helper cells and T peripheral helper cells) with B-cell activating capacities, in an effort to determine whether they are involved in the pathogenesis of T1D. Our preliminary results suggest that the activation of these cells occurs during the development of T1D, and therefore they could potentially be utilized as novel targets for immunotherapy of T1D and/or as biomarkers to evaluate the risk of disease progression.
