Although human and laboratory mice cancers have several common features, the mechanisms by which normal human and mouse cells are transformed to cancer cells are vastly different. The most distinct requirement between mouse and human cell transformation is requirement of inhibition of tumor suppressor PP2A for human, but not mouse cell transformation. PP2A regulates great number of mechanisms that regulate cellular responses to cancer drugs. Therefore, we hypothesize that human cancer cells are more therapy resistant than corresponding mouse cells because in human cells inhibited PP2A activates mechanisms that allow human cancer cells resist cancer therapies. Demonstration of the molecular basis of this difference could revolutionize our understanding of why many cancer therapy trials fail. Also, understanding of PP2A´s role in drug responses in mouse and human cancer cells could have important ramifications for development of future cancer therapies.
