Inflammatory diseases, often associated with pain or itch, are a major public health problem with significant unmet needs. Here, we focus on the role of TRPA1 in those indications. TRPA1 in known to be located in sensory neurons and, once activated by exogenous irritants, causes pain and itch. Based on our recent compelling findings, we aim to test the hypothesis that TRPA1 is an inducible gene expressed also in various non-neuronal cells and a significant endogenous mechanism mediating and amplifying inflammation especially in arthritis and (allergic contact) dermatitis. The project has significant potential to go beyond the state of the art: if successful, it will introduce a novel concept to target inflammation with simultaneous alleviation of inflammatory pain and itch through inhibition of TRPA1. Moreover, the project extends the current understanding on the general mechanisms how noxious environmental factors disturb homeostasis and trigger defense mechanisms in the human body.
