In sustainable biorefining concept all components of lignocellulosic material have to be utilised. Lignocellulosic biomass contains about 25% of hemicelluloses, which are composed of pentose sugar units that can be converted into biofuels, platform chemicals and novel materials by non-phosphorylated oxidative pathway of pentose sugars. Here we are studying the enzymes involved into this catabolic pathway of pentose sugars, especially pentonate dehydratases that are the bottleneck enzymes in the pathway. We are going to solve the crystal structures of pentose sugar modifying enzymes to understand the function of these enzymes at atomic level. Our aim is to get more efficient enzymes by protein engineering to improve the production of industrially relevant bio-based chemicals.
