LINE-1 retrotransposons are repetitive genetic elements that make up 17% of the human genome. A subset of them have can jump to new genomic locations via a copy-paste mechanism, referred to as retrotransposition. Retrotransposition in healthy cells is suppressed, but in many cancer genomes, LINE-1s are unleashed and contribute to genomic instability and heterogeneity. Additionally, when intragenic LINE-1s are de-repressed, transcription of the gene in which they reside can become misregulated. This transcriptional interference, along with retrotransposition, are two mechanisms whose functional contribution to tumorigenesis remains enigmatic. We will investigate the biological relevance of both processes in high-grade serous ovarian cancer, which is the most lethal type of gynecological cancer and where LINE-1 activity is known to be high. The molecular insights provided by this work could be utilized for earlier detection and improved therapeutic intervention in ovarian cancer.
