Extracellular-matrix (ECM) is an essential component of all multi/cellular organisms, it provides physical support for cells and dictates tissue-specific cell behaviour. A wide variety of ECM types exist, including basement membranes (BM), which are complex networks of proteins that cover cells. BM proteins are very poorly studied at the structural level as they are difficult to make due to them containing a large number of modifications. In this project we will apply synthetic biology for the efficient production of BM proteins to allow an unprecedented insight into structure-function relationships. This will allow molecular level understanding of normal physiological functions of the BM as well as of the progression of a wide-range of disease states. It will offer new insights into potential therapies to halt or reverse disease symptoms, including cancer progression and fibrosis.
