The nuclear lamina is a protein filament network that surrounds and protects the nucleus and thereby genetic material. Mutations in components of the nuclear lamina or lamina-associated proteins lead to degenerative diseases called laminopathies. Furthermore, changes in the nuclear lamina have been shown to be associated with physiological ageing. Thus, there is a critical research need to understand the mechanisms of tissue degeneration caused by alterations in the nucleus and the underlying chromatin. Strikingly, laminopathies commonly selectively impair tissues that experience mechanical strain, such as muscles and skin, through mechanisms that are unclear. We hypothesize that mechanical stress-induced remodeling of the perinuclear cytoskeleton, nuclear lamina, and heterochromatin are important mechanisms of mechanoprotection and maintenance of genome integrity. The current project aims to dissect the molecular mechanisms of these mechanoprotective functions.
