Every 13 seconds a new person is diagnosed with epilepsy. In 60% epileptogenesis is initiated by structural cause like traumatic brain injury (TBI). We have no treatments to alleviate epileptogenesis. The major obstacle in therapy development is the lack of biomarkers to identify patients at-risk of epileptogenesis. My objective is to validate sushi repeat-containing protein X-linked 2 (SRPX2) as a mechanistic biomarker for diagnosis of post-traumatic epilepsy. We will silence, stimulate, and injure neurons expressing SRPX2, and investigate the effect on plasma SRPX2 level using state-of-art technologies. We will assess samples from clinically relevant experimental model and humans with TBI. I consider this study highly original, feasible, and carrying a high-gain in case of a breakthrough in the discovery of SRPX2 as a mechanistic plasma biomarker for diagnosis of post-traumatic epileptogenesis, serving a large number of patients in-need of antiepileptogenic therapies after TBI.
