Epigenetic mechanisms control cell differentiation and malignant transformation, but their role in osteoblast differentiation is not fully understood. Development of modern molecular biologic methods has allowed rapid accumulation of epigenetic data. In our research program focusing on epigenetic control of osteoblast differentiation, we identified an epigenetic regulator, RCOR2, to be important for osteoblast differentiation. RCOR2 regulates the action of LSD1, which regulates cell differentiation but also cancer growth. We aim characterize RCOR2/LSD1 protein complex, their molecular mechanism of action in osteoblasts, and how depletion of these proteins affects bone metabolism in vivo. In addition, we will study the role of RCOR2 and LSD1 in osteo- and chondrosarcomas. Our studies will produce important new information on epigenetic mechanisms in bone and may identify novel drug targets for bone-related diseases.
