The delicate balance between viral infection and host response is controlled by a multitude of interactions between molecules from both the host and the virus. We propose to study pathogenic RNA virus-host interactions. Significantly, we will act on two of our major breakthroughs: The first is that we have identified a new class of molecules that interact with picornavirus capsids and prevent infection. The second is our new assembly model for the human parechovirus where we have shown multiple, identifiable packaging sequences within the HPeV genome responsible for virus assembly (Shakeel et al Nature Comm, 2016 & 2017). We have two main objectives: 1) to understand the mode of binding for the new capsid binders and to investigate their applicability as broad range inhibitors of picornaviruses, 2) to explore the role of conformational change and protease activity on HPeV assembly. Thus, we will use structural cell biology and computational science to innovate new products.
