Amygdala is known to be important for primary emotions, such as fear. Additionally, it is known that the central nucleus of amygdala is involved in pain processing. The role of amygdala in the development of neuropathic symptoms is still poorly known. In the proposed study, we characterize the role of amygdala and its plasticity in the development of neuropathic pain hypersensitivity and comorbid affective-cognitive disorders. This includes determining whether neuropathy-associated structural and functional plasticity is causally related to neuropathic symptoms. Using electrophysiological methods, we determine the types of neurons and their interactions in the neural circuitry originating in the amygdala and maintaining neuropathic pain hypersensitivity. The results are expected to increase understanding of mechanisms underlying neuropathy and thereby, the results may provide possibilities for developing diagnostic and mechanism-based therapeutic methods of neuropathy.
