Mitochondria play an important role in cellular respiration. We study the process of mitochondrial fatty acid synthesis (mtFAS), which is essential for respiration and indispensable for human viability. MtFAS is required to make lipoic acid, an important mitochondrial enzyme cofactor. We have recently shown that mtFAS also generates other yet unknown products which are required for the maintenance of healthy mitochondria and respiratory activity and necessary for the ability of mitochondria to respond to metabolic changes. Deficiencies in mtFAS lead to severe diseases. Using mouse, cell culture and yeast as study objects, we will characterize the components and products of mtFAS, the mechanisms how these products control mitochondria and model disorders that are caused by mtFAS deficiencies. Our work in these models will help our understanding of the role of mtFAS in cellular metabolism, health and disease and ultimately aid in finding treatment for mtFAS disorders.
