Project description
The project will elucidate how genome structure and a metabolic network cooperatively regulate adaptive evolution. The co-regulation influences all proliferating cells, from aging to tumor cells, and to developing stable industrial biotechnology hosts. In this project, yeast variant libraries are engineered using synthetic biology tools by altering either the genome structure or the metabolic network. Thereby their contributions can be dissected in adaptive laboratory evolution (ALE). The ALE experiments will be performed in conditions designed using genome-scale metabolic model simulations for exposing the variant effects on adaptive potentials. The adaptive potentials of the variant strains will be assessed and next-generation sequencing will be used to identify de novo mutations revealing the interplay of genome structure and metabolic network in regulating adaptive evolution. This way the project will provide a substantial leap towards predicting adaptive evolution.
