Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disease worldwide and causes mortality related to liver and heart diseases. NAFLD is a common name for continuum of progressive liver abnormalities starting from lipid accumulation to liver, continuing with the process of inflammation and sometimes leading to irreversible fibrosis and life-threatening cirrhosis and liver cancer. NAFLD is closely associated with and shares risk factors with type 2 diabetes and other components of the metabolic syndrome; however, exposure to environmental contaminants, the so-called endocrine-disrupting chemicals, represent an addition etiological factor. In this project, we investigate the role nuclear receptor Pregnane X receptor (PXR) in the pathology and progression of NAFLD induced or aggravated by chemical exposure.
