Project description
Magnetic resonance methods, NMR spectroscopy and MRI, are versatile tools for chemical and biomolecular analysis. Their major limitation concerns the low thermal magnetization of samples, which leads to low signal amplitudes. The magnetization can be improved by orders of magnitude with spin hyperpolarization. The methods using parahydrogen as a source of hyperpolarization are the easiest to implement with inexpensive equipment. However, their potential for biomolecular studies is far from the application since the required chemical activation of parahydrogen needs the use of toxic metal-containing catalysts. In this project, we develop metal-free activators for parahydrogen to create hypersensitive non-toxic biosensors for NMR and MRI studies. The biosensors have affinity to biomolecules, and provide very strong NMR response due to the hyperpolarization. This allows detecting target molecules, providing means for biomolecular analysis, imaging and drug delivery monitoring.
