Project description
One of the hallmarks of cancer is deregulated extracellular matrix dynamics. This proposal is motivated by our findings that expression of type XVIII collagen (ColXVIII), a ubiquitous extracellular component discovered by us, significantly increases in malignant cancer. We hypothesize that ColXVIII interacts with growth factor and cell adhesion receptors to coordinate their cooperative signalling and thereby amplify cell regulatory signalling in cancers. Here we propose to expand our data on expression of ColXVIII in human cancers, study the significance of ColXVIII in mouse carcinoma models, identify mechanisms whereby ColXVIII affects epithelial cell adhesion and signalling, and conduct studies to assess the eligibility of ColXVIII as a drug target in human cancers. We expect breakthroughs in understanding the complex regulation of tumourigenesis involving ColXVIII, and the results may have high medical relevance and potential for novel therapies.
