Project description
Proper oxygen levels have a vital importance for the normal function of cells and tissues. The response of cells to hypoxia is mediated by the hypoxia-inducible transcription factor (HIF). Hydroxylation of specific prolines by HIF prolyl 4-hydroxylases targets HIF for rapid proteasomal degradation in normoxia. In hypoxia, this hydroxylation is inhibited, HIF escapes degradation and induces numerous hypoxia-regulated genes. Detailed information on mechanistic aspects of the hypoxia response and its key enzymes will have a major impact in furthering our understanding of how cells and tissues survive and respond to situations of acute or chronic hypoxia. Such information will create novel opportunities for pharmacologic interventions in pathological conditions characterized by insufficient oxygen levels.
