Project description
Degradation of fatty acids is an important source of energy in a variety of organisms. The purpose of this project is to understand the molecular properties of one of the multifunctional enzymes responsible for degrading fatty acids, known as ‘Trifunctional Enzyme’ (TFE). TFE catalyses three reactions and it is assembled as an A2B2-tetramer. In human, defects in the functioning of this enzyme leads to sudden infant death and many other health problems including heart failure. To understand the functioning of this enzyme complex we will determine the structure of human mitochondrial TFE and a related TFE from E. coli. Another major question concerns the substrate specificiy of the Mycobacterium tuberculosis TFE which will provide insight into the function of this enzyme and which has potential in drug discovery applications. Mechanism of channeling of the substrates from one active site to the other in these multi functional enzymes will also be studied in great detail.
