Project description
It is known that individual’s DNA methylation status changes during one’s lifetime and methylation changes can be also inherited. However, it remains largely unknown what affects the human methylome and to what extent and if the epigenome is heritable. The primary aim of our study is to research lifelong stability and heritability of DNA methylation and microRNA expression profiles in more than 30-year long Young Finns follow-up and its offspring studies. DNA methylation and microRNA expression profiling will be performed to a subpopulation of the study participants at three time points (T1=ages 9-24, T2=ages 24-39 and T3=ages 34-49). We will focus on the lifetime trajectories of indicators associated with metabolic syndrome and their impact on epigenetic changes and development of prediabetes and diabetes as well as CVD and CVD mortality. In addition we will provide information about the biological function of these epigenetic profiles using induced pluripotent stem cells (iPSCs).
