Project description
G protein-coupled receptors (GPCRs) are polytopic membrane proteins. They play a fundamental role in mediating extracellular signals into cellular responses, and they are the major targets of pharmaceuticals. However, very little information is available on how GPCRs acquire the correct conformation during biosynthesis and are transported to the cell surface, or how their functional activity is regulated. This project aimed to explore previously unknown mechanisms that regulate GPCR expression, using model wild-type and mutant receptors, and a combination of molecular biological, biochemical and cell biological approaches. The results provided new insights into the mechanisms involved in GPCR folding, maturation and trafficking. Improving or correcting defective protein folding and/or trafficking of mutant GPCRs and regulating expression of wild-type receptor levels at the cell surface may represent a major advance toward the development of new therapeutics.
